Thank for the last comment @crythias, it triggered a thought about the SMTP relay on my new Exchange 2010 server, I amended the persmissions on the Authentication tab and it's now working as normal.

I was only using IMAP as that was what worked in the first place, I don't mind not keeping the emails.
