Have few questions on ACL.
To start - do I need to restart httpd service to get ACL defined in Config.pm to come into effect ?
OTRS Version: 3.3.1
OS: Cent OS 6

Usually not. Unless there's some big hiccup or you're using some cacheing. Check your logs.
If nothing's crashing, then it's possible your filter is doing what it's told, not necessarily what you want it to do.
