Is the file existing, is it configured via sysconfig? Is it intented to use it?

Yes I have the file in location

/otrs/bin/cgi-bin/rpc.pl

Also check apache error log and OTRS log

ans for q2. copied directly to /otrs/bin/cgi-bin/ folder
ans for q3. yes we need to use because we are on boarding customer form other software
